rhErbB3-f is a recombinant human ErbB3 receptor fragment produced by expression in E. coli. After purification and formulation with adjuvant, it can be used for immunization to induce the production of specific anti-ErbB3 antibodies for the treatment of patient with breast cancer characterized by the overexpression of ErbB2. rhErbB3-f, therefore, is as a therapeutic cancer vaccine.

| Pharmacological Action |
| Results of animal experimentation have shown that vaccination with rhErbB3-f delays the occurrence of spontaneous breast cancer tumors in FVB/N neu transgenic mice and inhibits the development of the tumors, effectively arresting the growth of in situ transplanted breast in this animal model. Moreover, anti-rhErbB3-f polyclonal antibodies can directly inhibit transplanted breast tumors in nude mice and inhibit in vitro cell growth. Vaccination of mice with recombinant mouse ErbB3 fragment also inhibits the growth of in situ transplanted breast cancer cells in FVB/N neu transgenic mice, indicating that immune tolerance can be overcome. |
Thus, the main pharmacological action is through the induction of specific anti-ErbB3 antibodies, which effect tumor cell growth inhibition. |
Safety |
No adverse side effects or autoimmune responses associated with the drug have been observed in detailed safety pharmacology, acute toxicity, and long term toxicity studies in mice or monkeys.
